Research Overview
● Immunization Technique for Induction of Antibodies Against Membrane Proteins Is a Bottleneck for Development of Antibody Drugs
With the rapid expansion of antibody drug development, the antibody market is growing, and demand for antibody reagents in basic research is also increasing. In basic research, there is strong demand for antibodies that recognize specific cell types, and most antibodies used in flow cytometry and related techniques target membrane proteins. However, conventional immunization technique faces a major challenge in efficiently generating antibodies that recognize the higher-order structures of membrane proteins.

● Development of a Novel Immunization Technique Using Spleen-Targeted DDS Technology for Induction of Antibodies Against Membrane Proteins
We have developed a spleen-targeted immunization technology that delivers antigens to the spleen using liposomes. Compared with the conventional subcutaneous immunization method, this technology enables a shorter immunization period, induction of highly diverse antibody repertoires, multiple IgG subclasses, and high-affinity antibodies. Furthermore, by preparing liposomes, incorporating membrane proteins and applying spleen-targeted immunization, we successfully induced antibodies against membrane proteins that could not be obtained using the conventional method.

Potential Applications and Commercialization Prospects
● Creation of Novel Antibody Drugs and Antibody Products Targeting Membrane Proteins
Antibodies were generated and evaluated by spleen-targeted immunization using liposomes incorporating membrane proteins. This approach successfully induced antibody populations with high specificity for cell membrane proteins. Proteomic analysis was conducted to identify the membrane proteins recognized by these antibodies, revealing antibody repertoires against membrane proteins that cannot be obtained using conventional immunization technologies.
This method is expected to contribute to antibody drug discovery targeting novel membrane protein antigens, as well as to the development of next-generation antibody therapeutics with performance superior to that of existing antibody drugs.
