Bio RI共同利用実験施設土田助教授の参画を得て、共同研究を展開してきた。


1.アクチビンの生理作用とその制御機構

TGF-bファミリーに属するアクチビンのシグナル伝達系の細胞内外における制御機構を解明することにより、臓器形成の素過程への理解を深め、先端的再生医療への展開を目指している。


  1. アクチビン受容体細胞内ドメインと相互作用するタンパク質因子を探索して、PDZドメインを持つARIP(Activin Receptor Interacting Protein)分子群を発見した。
    5個のPDZドメインを有するARIP1は神経シナプスに高発現しており、アクチビン受容体に加え、NMDA受容体とも結合して、これら膜タンパク質の足場タンパク質としての役割を果たしていることを見いだした。ラット海馬細胞培養系を用いて長期記憶におけるアクチビンの役割を検討した。後シナプス肥厚(PSD)に存在するARIP1上でアクチビンのシグナルがアクチビン受容体/Fynを介してNMDA受容体に伝達されてCa2+の流入が顕著に促進されること、このアクチビンによるNMDA受容体の活性化が24時間にわたって認められることを観察した。この結果は、海馬においてアクチビンの長期記憶ポテンシエーターとしての役割を強く示唆している。
    1個のPDZドメインしか持たないARIP2やそのスプライシング産物ARIPzipは、G-タンパク質、Ral-Bp1やRab5Cともそれぞれ結合してアクチビン受容体の細胞膜上での動態を調節していることを明らかにした。
  2. 新規アクチビン結合タンパク質FLRGを発見し、それがアクチビン結合能を持つタンパク質として既に見いだしていたフォリスタチンと同様、アクチビンに加えて筋細胞分化抑制因子マイオスタチンとも強く結合し、その作用を阻害することを明らかにした。
    マイオスタチンの作用を選択的に阻害するフォリスタチン構造変異体を調製することに成功し、その筋組織特異的発現トランスジェニックマウスを作製した。この遺伝子改変マウスは、筋肉量が著増し、筋ジストロフィーモデルマウスとの交配実験により、その病態が軽減することを明らかにした。また、FLRGのトランスジェニックマウスも同様なフェノタイプを示したことから、これらタンパク質因子が難病に指定されている筋ジストロフィーをはじめ各種筋肉疾患に対する新しいタイプの治療法への応用が示唆された。

2.幹細胞の分化制御とプロテオーム解析

幹細胞の未分化性、多能生、自己複製性などの自己管理制御システムの全貌を解明して、幹細胞を用いる再生医療の抱える問題点を明確にすることを目的とする。


  1. 幹細胞及びその分化過程において特異的に機能するタンパク質をプロテオーム解析により系統的かつ網羅的に同定するという新しい試みを開始した。LIF存在下及び非存在下におけるマウスES細胞のタンパク質発現プロファイルをマス分析により作成した。その結果、ES細胞の未分化状態ではHMG-2、AOF2、SWI/SNFなどのクロマチン関連タンパク質がメジャーなタンパク質として同定された。幹細胞の未分化性維持にエピジェネティクス制御の重要性が示唆された。

3.神経系におけるイオン環境恒常性維持機構

神経活動が活発な神経系(脳)においていかに神経細胞内外のイオン環境の恒常性が保たれるのか、病態ではどのように変わるのか研究している。頭蓋骨で囲まれた脳の2/3を占めるグリア細胞に、虚血などの原因によりイオン輸送機能の異常が起きた時、グリア細胞の膨潤が起こり、神経とグリア間の間隙が狭まり、イオン環境の恒常性が崩れ活動電位伝播の阻害に至る。局所血流量の上昇によるグリア細胞のボリュームコントロール、それによる速やかな神経活動機能回復の実例を検証中である。
